趋化因子网络在肿瘤微环境免疫应答中的作用

趋化因子(Chemokines)是一类由细胞分泌的小细胞因子或信号蛋白,根据N端保守的半胱氨酸(C)的数量及排列方式,可将趋化因子分为四类:C、CC、CXC和CX3C。这些趋化因子可以通过细胞表面G蛋白偶联的七次跨膜趋化因子受体发出信号,从而刺激细胞定向趋化迁移。稳态趋化因子在生理条件下组成性表达并在细胞迁移和归巢中发挥作用,而炎性趋化因子会在炎症部位迅速分泌,从而将效应细胞募集到发炎组织中。因此,趋化因子在免疫系统的发育和稳态中起着核心作用,参与促进免疫细胞的激活、分化、增殖和凋亡并参与所有保护性或破坏性免疫和炎症反应。
同时,趋化因子信号传导和各种细胞群的趋化性在肿瘤微环境(TME)中也起着核心作用。在TME中,免疫细胞、肿瘤细胞和肿瘤相关细胞都会释放多种趋化因子,这些趋化因子可以随时空变化,吸引不同类型的促肿瘤和抗肿瘤免疫细胞。免疫细胞的迁移会进一步影响肿瘤的进展和转移,并直接影响免疫反应。研究前沿揭示,趋化因子具有影响抗肿瘤免疫反应和肿瘤进展的多效性功能。考虑到这一点,在过去十年中已经建立了利用或靶向趋化因子的治疗策略,从而在临床试验中测试了许多新的候选药物。值得注意的是,在基于趋化因子的疗法中,趋化因子配体与受体结合高度重叠,同一受体可与不同的趋化因子结合,反之亦然。因此,只有同时抑制具有相同生理功能的趋化因子及其受体,才能达到更健康、更持久的治疗效果。
