Human RANK (Luc) HEK293 Reporter Cell

描述(Description)

The Human RANK (Luc) HEK293 Reporter Cell was engineered to express NF-kB signaling response element driving luciferase expressing systems. When stimulated with human RANKL protein, the RANK/RANKL interaction drives NF-kB-mediated luminescence. Blocking the RANK/RANKL interaction by either anti-RANK or anti-RANKL antibodies results in a decrease in luminescence.

Application

RANK APPLICATION

Inhibition of human RANKL protein-induced reporter activity by anti-human RANKL neutralizing antibody.
This reporter cell was incubated with serial dilutions of antibodies in the presence of human RANKL protein (Cat.No.RAL-H5240) with a final concentration of 0.3 μg/mL. The EC50 of anti-human RANKL neutralizing antibody is approximately 0.1742 μg/mL.

背景(Background)

RANK / TNFRSF11A is a member of the tumor necrosis factor receptor family. RANK / TNFRSF11A shares significant amino acid homology with other members of the TNF R family in its extracellular four cysteine-rich repeats. Functional genomics in mice showed that RANKL (TNFSF11), a member of the TNF ligand superfamily, and RANK (TNFRSF11a), the cognate TNFR family receptor for RANKL, were essential for osteoclastogenesis in vivo. A functional interaction between RANKL, expressed by bone stromal cells of the osteoblast lineage, and RANK, expressed by osteoclast precursors of hematopoietic myeloid lineage, is necessary for osteoclast differentiation, survival, and activation.
