Our Recombinant Human FGF basic Superior Stable Mutant Protein, developed via an AI-modified and experimentally validated closed-loop efficient protein design platform, is the optimal FGF2 mutant with superior thermal stability and activity compared to wild-type FGF2, making it more suitable for various stem cell culture processes. This reduces medium replacement frequency, lowers contamination risks from frequent operations, and saves costs/time for stem cell culture by minimizing reagent consumption and labor, especially in large-scale applications.
It is produced under our rigorous quality control system that incorporates a comprehensive set of tests including sterility and endotoxin tests. Product performance is carefully validated and tested for compatibility for cell culture use or any other applications in the early preclinical stage. Recombinant Human FGF basic Superior Stable Mutant Protein Induces Proliferation of NIH-3T3 Mouse Fibroblast Cells. (A) Human FGF basic Superior Stable Mutant Protein, premium grade (Cat. No. BFF-H5113) or (B) wild-type (WT) Human FGF basic Protein, premium grade (Cat. No. BFF-H4117), were either untreated or incubated in culture media at 37°C for 3 days. Following 3 days of treatment at 37°C, Recombinant Human FGF basic Superior Stable Mutant Protein retained activity comparable to the untreated heat-stable variant, demonstrating its enhanced thermal stability. In contrast, the WT Recombinant Human FGF basic Protein exhibited significant activity loss, indicating inferior thermal stability.

100 ng/mL Superior Stability for Recombinant Human FGF basic Superior Stable Mutant Protein (Cat. No. BFF-H5113) or (B) wild-type (WT) Human FGF basic Protein, premium grade (Cat. No. BFF-H4117), were incubated in culture media at 37°C for 0h, 8h, 24h, 48h and 72h. The concentration of residual FGF2 was detected by resDetect™ Human FGF2 ELISA Kit (Residue Testing) (Cat. No. RES-A009).
Following 72h of treatment at 37°C, Recombinant Human FGF basic Superior Stable Mutant Protein retained higher concentration comparable to the WT FGF2 protein, demonstrating its enhanced thermal stability. In contrast, the WT Recombinant Human FGF basic Protein exhibited significant concentration loss, indicating inferior thermal stability.

The specific activity of Human FGF basic Superior Stable Mutant Protein, premium grade is > 1.60 x 10^7 IU/mg, which is calibrated against Basic Fibroblast Growth Factor WHO International Standard (NIBSC code: 90/712) (QC tested).

活性(Bioactivity)-Stem Cell Culture

100 ng/mL of either Recombinant Human FGF basic Superior Stable Mutant Protein (Cat. No. BFF-H5113, designated as HS-FGF2) or wild-type (WT) Human FGF basic Protein of premium grade (Cat. No. BFF-H4117, designated as WT-FGF2) was added to the iPSC culture system. The culture protocol involved daily medium replacement or a reduced feeding schedule with medium changes every 2 days. Cell counts were assessed via AO/PI staining from passage 1 to passage 3.
The results indicated that the Recombinant Human FGF basic Superior Stable Mutant Protein significantly promoted stem cell proliferation, maintaining high cell numbers even under both daily feeding and the reduced 2-day feeding regimen, with performance comparable to that of competitor T. In contrast, the WT Recombinant Human FGF basic Protein showed a marked decrease in cell numbers under the reduced 2-day feeding condition (Data is the average of 3 independent experiments, error bars ± SD).
