I have been using the unconjugated FMC63 for a while and as soon as we knew about the conjugated FMC63 we wanted to use this as it will be saving us 1.5 hours in the staining procedure. We used this antibody and we were able to see a better seperation of positive and negative poulations. Its easy to use and 1:25 dilution works really well. I will recommend this 200%
We are a CAR-T therapy research group and we use FITC anti-FMC63 and FITC-Labeled Human CD19 protein to detect CAR19 cells both in patient blood and in vitro cultures by flow cytometry. The results obtained so far with both reagents are reliable and reproducible, with a clear and easy detection of positive population even when reagents are directly incubated with blood samples.
FMC63 is an IgG2a mouse monoclonal antibody specific for CD19, which is a target for the immunotherapy of B lineage leukaemias and lymphomas. FMC63 scFv is the most commonly used ectodomain component of CD19-specific CARs. So far, most of reported CART19 trials contain the anti-CD19 scFv derived from FMC63, including the two FDA-approved CARs Kymriah and Yescarta.
